Research Interests
Developing new methods for comprehensive glycoproteomic studies, as well as applying glycoproteomic methods into various biomedical samples with different physiological and pathological conditions for a systems biology-level understanding of the consequences of perturbed biological processes.
During the last few years, he mainly developed:1)StrucGP, a modularization strategy together with an algorithm, for de novo interpretation of N-glycan structures on intact glycopeptides using tandem mass spectrometry (Nature Methods, 2021); StrucGP can discriminate glycan isoforms on intact glycopeptides and facilitate the identification of rare/new glycan structures in different biomedical samples.2)the NGAG method and related tools for comprehensive characterization of N-glycoproteome, with which information of N-glycans, glycoproteins, and glycosites, as well as site-specific glycosylation and occupancy can be obtained simultaneously (Nature Biotechnology, 2016);3)new methods for characterizing site-specific N-glycans with certain structures (Analytical Chemistry, 2019); and4)Real-world application of glycoproteomics and the successful discovery of glycosylation alterations in biomedical samples including ovarian cancers (Cell, 2016; Nature Communications, 2020) and liver cancer (Theranostics, 2021). These applications are crucial for the understanding the biological roles of glycosylation and could help us to explore the possibilities of glycans as diagnostic biomarkers and therapeutic targets.
